Tissue Factor expression on monocytes

I am trying to set up an assay for essessing TF expression on human
monocytes as a determinant of PCA. I will be obtaining my PBMC from whole
blood (probably sodium heparinized). I am wondering if anyone has
experience in this area and could help me set up the protocol? I am
thinking of stimulating the PBMC in OPTI-MEM/antibiotics/10%FCS/1X
B-mercaptoethanol with PMA and ionomycin for four hours prior to staing. Is
this appropriate? Should I block protein synthesis with Brefeldin A and
stain for intracellular TF as well? Any help would be appreciated. Thanks
