Optical disks etc ..

Hi,
In few comments regarding recent questions ...
Optical disks:==> We are using a Sony MO disk unit (NWP-559) and
disks supplied by 3M. Works finebut once every couple of months or so
you get a bad sector on the disk. This unit is attached to a PC, and the
only way to fix these bad sectors is using the propriety software, not
the operating system software. Apart from this it's good.
7AAD:==> Got around ten different people here using 7AAD on mostly
mouse lymphocytes, and can't say that we have seen the problems you describe
. The main reason it's used here is not the compensation, it is the not
having to use RNAase that people like.
On a different note, there is currently a project underway here to
sex birds, based on PI staining. The problem we have encountered is that the
peaks on PI histogram drift up the longer the sample is on the instrument. We
have tried running them after 5minutes, 1 hour, 6 hours, and 24 hours of
staining, but the same thing occurs. Interestingly this occurs to a lesser
extent on the FACStar PLUS than the 'Scan, as I initially thought this may be
due to a free dye being taken up over time, or bleeding into the sample lines
but his appears not to be the case. Any suggestions regarding this would be
appreciated.
