In this experiment, rat T cells were loaded with the green Calcium indicator Fluo-3 . Dissolution of the dye in aqueous solution was facilitated by the use of Pluronic f-127, a non-ionic detergent. A modest calcium flux was noted in only one of three animals in response to the cytokine, Rantes. Optimal loading and maximum Ca2+ flux was verified by the addition of a calcium ionophore , Ionomycin, seen in the lower right dotplot
