Dr. Matthias Haury inquires about flow cytometric techniques
discriminating apoptotic cells. We have recently published the
methodological chapter (Methods in Cell Biology, 41, 15-38, 1994)
presenting 10 different flow cytometric methods to identify dead
cells and analyze the mechanisms of their death (apoptosis vs.
necrosis). Among these techniques there is a description of the DNA
strand break labeling in apoptotic cells using exogenous terminal
transferase, developed in our laboratory (Gorczyca et al., Int. J.
Oncol., 1, 639, 1992), which provided the basis for the ONCOR kit. I
have already presented on this forum (February?) the simplified,
single step DNA strand break labeling protocol, which will be
described in the forthcoming issue of CYTOMETRY. The single step DNA
strand break labeling kit soon (next month ?) will be provided by
Phoenix Inc., (San Diego).
Two other methodological chapters on analysis of apoptosis by flow
cytometry which will be published this year.
