Dear flowers,
since many months we are trying to get a cyclin D1 signal in
parralell to DNA staining in bladder cell lines. Immunocytochemistry
of cultured cells and even staining in paraffin sections works
nicely, however all the protocols we have tested and modified
extensively ( Paraformaldehyd/various soaps as Tween , Nonidet etc)
and mixtures of methanol and ethanol do not result in a specific
staining pattern for cyclin D1. We are using commercial antibodies from
Pharmingen, and they advertise with nice dot blots of the cyclin
antibodies. Does anybody have a working protocol on epithelial cell
suspensions?
We would be grateful for a little help, we are routinely staining
Ki67, p120 and other nuclear antigens with success, and would so much
like to get this going as well.
