We recently embarked on an alveolar macrophage project, staining for surface
molecules. (Fun, fun, fun) As perviously suggested by the kind members here,
the autofluorescence of these cells is somewhat of a pain. However I found
some papers that could help others who face similar problems in overcoming
them. One paper actually uses the autofluorescence to an advantage.
"Multiparameter flow cytometric analysis of inflammatory cells contained in
bronchoalveolar lavage fluid. J.Imm.Meth. 172(1994) 59-70"
"Application of a flow cytometric method using autofluorescence and tandem
fluorescent dye to analyse human alveolar macrophage surface marker.
J.Imm.Meth. 172(1994) 17-24"
We are using this tandem dye (R-PE-Cy5 - excitation 488nm emission
660+-20nm). Has anyone come across flow standard beads that will enable us
to quantitate fluorescence (MESF) at this emission wavelength. I have looked
in the FCSC catalog but no banana.
Fanx in advance
